If you have all perks, the 4th perk will have its modifier activated.
From cod wiki for quick revive: “When placed in the modifier slot, it is upgraded so that when reviving a teammate, both players are instantly healed to maximum health, and both players get a speed boost. When in the modifier slot, the player will also get a speed boost any time they regenerate health.”

Whatever perk is in your Tonic slot will glow once all other perks have been bought. It basically adds an extra improved effect to the perk itself. If you look at the perk screen in the main menu, it gives you a description of what each Tonic upgrade does for each Perk.
